Reactive Oxygen Intermediate Production of Phagocytes from Human Cord Blood

Abstract

It is well known that many aspects of neonatal immune function are immature, including B and T cells functions and granulocyte and macrophage functions. Therefore, newborns have increased infection, but decreased febrile response to infection. In
this
study, the authors tried to measure the capacities of reactive oxygen intermediate(ROIs) production of phagocyte from newborns and adults.
1. There were decreased phagocytic activities in the cord blood phagocytes compared with the adult peripheral blood phagocytes.
2. There were increased productions of ROIs in the cord blood monocytes compared with the adult peripheral blood monocytes.
3. There were decreased productions of ROIs in the cord blood polymorphonuclear leukocytes (PMNL) compared with the adult peripheral blood polymorphonuclear leukocytes.
4. In PMNL of cord blood, the change of intracllular calcium was the same pattern as PMNL of peripheral blood when the cells stimulated with phorbol 12-myristate 13-actetate(PMA0 after incubating Fluo-3AM.
The above results suggest that phagocytes of newborns might be at the different maturation stage and under the control of different levels of cytokines.
